Clinical Rationale & Mechanism of Action 1. Berberine as a Bioactive Alkaloid Berberine is a potent plant alkaloid with broad-spectrum antimicrobial, anti-inflammatory, and gastrointestinal regulatory effects. Found in high concentrations in barberry, Oregon grape, and goldenseal, it exerts its effects by influencing mucosal integrity, ion transport, motility, and microbial adhesion. 2. Gastrointestinal Barrier Protection Berberine supports epithelial tight junction formation and reduces intestinal permeability, thereby enhancing gut barrier function and mitigating "leaky gut" symptoms.⁵ 3. Anti-Diarrheal Mechanism Modulates ion and water transport across epithelial cells by upregulating Na⁺/H⁺ exchangers and aquaporins, reducing excess water in the bowel and helping to manage diarrhea.³˒⁴ 4.
